This issue of the Journal contains three articles about theadverse cardiovascular effects of agents that selectively inhibitone form of prostaglandin endoperoxide synthase, commonly knownas cyclooxygenase-2 (COX-2).1,2,3 This is part of a long bench-to-bedsidestory with an adverse outcome that was not widely anticipatedat its start. Unfortunately, as the evidence began to suggestunexpected toxicity of this group of agents, the same zeal thathad driven the clinical investigation to show their gastrointestinalsafety was not evidenced by studies designed to show their cardiovascularsafety.
